主要从事化学与酶法定向合成海洋来源糖类化合物和糖胺聚糖类寡糖,开发新颖的糖聚合物合成方法并应用于活性多糖模拟物的制备,特征糖类化合物的活性评价及糖类药物研发工作。近年来在Chem. Rev., Nat. Chem. Biol., ACS Macro Lett., Biomacromolecules等杂志发表SCI论文50余篇。主持国家自然科学基金、山东省重点研发计划以及参与国家重点研发计划、国家重大新药创制等多项国家及省部级课题。 | ||||

糖胺聚糖(GAGs)是一类线性多糖,由硫酸化的二糖重复单元组成,参与多种生命过程。乙酰肝素酶是一种内源性β-D-葡萄糖醛酸苷酶,在人类肿瘤细胞和特定组织(例如哺乳动物的胎盘)中高水平表达。乙酰肝素酶与多种生长因子的激活有关,这些因子在导致组织修复、细胞粘附、生长、增殖和分化、癌症血管生成和炎症等相关疾病中发挥着重要作用。最近,我们课题组通过正交脱保护化学合成了区域选择性硫酸化的半乳糖胺衍生物,并基于开环易位聚合(ROMP)和后修饰策略生成了系列糖聚合物。具有多种硫酸化模式的半乳糖胺聚合物显示出对乙酰肝素酶的显着抑制作用,其中含有3,4,6-O-硫酸化的GalNAc的糖聚合物能够有效抑制乙酰肝素酶活性以及肿瘤细胞的增殖。在此基础上,具有肿瘤靶向作用的iRGD肽通过末端功能修饰引入。流式细胞术和共聚焦显微镜结果显示,末端功能化的糖聚合物,可以有效进入HeLa细胞并抑制参与肿瘤细胞增殖的信号通路。这些发现有利于推动针对特定肿瘤靶向疗法的新型糖模拟物药物开发。 本课题组通过自由基聚合(FRP)与化学酶法合成联用技术,首次建立了高效制备结构明确的HS聚合物的反应体系,获得了系列侧链长度可控和不同硫酸化修饰的HS聚合物。采用表面等离子体共振(SPR)技术,研究了HS 糖聚合物与晚期糖基化终产物受体(RAGE)的相互作用,发现全硫酸化的HS三糖聚合物与RAGE 蛋白具有最强的结合活性,其KD值为0.3 nM,具有开发成RAGE蛋白拮抗剂的潜力。 | ||||
